高糖对人肾小球系膜细胞细胞间黏附分子-1及层黏连蛋白表达的影响 被引量:3

摘要 目的探讨高糖对人肾小球系膜细胞表达细胞间黏附分子-1(intercellular adhesion molecule-1,I-CAM-1)及层黏连蛋白(laminin,LN)的影响。方法将培养的人肾小球系膜细胞分为正常糖对照组、高糖组和甘露醇对照组,分别在24、48、72h收集培养液上清,采用ELISA法检测上清液中ICAM-1和LN的表达。结果人肾小球系膜细胞表达ICAM-1在24h开始升高,48h达高峰,72h下降。LN表达呈时间依赖性。且高糖组ICAM-1及LN的表达显著高于正常糖对照组及甘露醇组(P<0.05),正常糖对照组与甘露醇组对比ICAM-1及LN的表达差异无显著性(P>0.05)。结论高糖可导致人肾小球系膜细胞ICAM-1及LN表达增加。I-CAM-1对肾脏可能具有炎性损伤作用,LN的增加表明肾小球系膜外基质的过度表达,二者均可促进糖尿病肾病的发生发展。从而为糖尿病肾病的治疗提供了新靶点。 【Objective】 To investigate the effect of high glucose on the secretion of intercellular adhesion molecule-1(ICAM-1)and laminin (LN) in the human mesangial cells.【Method】Human mesangial cells were cultured in vitro and divided into nomal glucose group, high glucose group and mannitol group. Supernatant were collected at 24 h, 48 h and 72 h. The levels of ICAM-1 and LN in the supernatant were detected by ELISA assay. 【Result】The level of ICAM-1 was increased at 24 h, reached peak at 48 h and decreased at 72 h. The level of LN was time dependent. Compared with nomal glucose group and mannitol group, levels of ICAM-1 and LN were significantly higher in high glucose group (P 0.05). Levels of ICAM-1 and LN were no significant difference between nomal glucose group and mannitol group (P 0.05).【Conclusion】High glucose could increase the secretion of ICAM1 and LN in human mesengial cells. Kidney could be injured by inflammatory mechanism of ICAM-1 and the increasing of LN indicated the over-expression of extracellular matrix. Both could cause the development and progression of diabetic nephropathy. It also provided a new therapy target of diabetic nephropathy.
