摘要
目的观察蛋白激酶C-α(PKC-α)与热休克蛋白HSP70在兔自体静脉移植中的表达的动态变化,探讨其与移植后血管内膜增生的可能关系。方法采用兔自体颈静脉移植模型,25只新西兰大白兔随机分为5组,分别于术后3、7、14及28d取材,应用Real-Time PCR法、免疫组织化学法分别检测各组中PKC-α、HSP70 mRNA与蛋白的表达变化。结果PKC-α、HSP70 mRNA在术后3d时表达升高,而后渐降至正常,两者变化呈正相关(r=0.967,P<0.01),蛋白检测表达变化与其一致。结论静脉移植后可能通过PKC-α信号传导通路使热休克蛋白HSP70表达升高,与促进血管平滑肌细胞增殖有关。
【Objective】To observe the expression of PKC-α and HSP70 in autogenous vein graft and their relationship with intimal hyperplasia.【Methods】Twenty-five New Zealand rabbits were randomly divided into five groups.The right common jugular vein was transplanted to the contralateral common artery.The specimen was collected at 3,7,14,28 d after operation.In each group,the PKC-α and HSP70 mRNA were detected by Real-Time PCR.Immunohistochemistry was used to detect the expression of the PKC-α and HSP70.【Results】Compared with SO group,the expression of PKC-α and HSP70 mRNA and protein increased significantly at 3 d after operation,then returned to normal gradually.【Conclusion】The PKC-α was involved in the expression of the HSP70 in the autogenous vein graft,which was related to proliferation of the vascular smooth muscle cell.
