摘要
比较五种染色方法对小鼠圆形精子(round spermatid,ROS)染色的效果,寻找一种鉴定圆形精子简单、有效的染色方法。以性成熟的昆明白鼠为实验材料,制备睾丸细胞悬液,分别用瑞氏、瑞氏-姬姆萨、姬姆萨、改良品红、苏木精五种方法对小鼠生精细胞进行染色,比较不同染色方法下圆形精子的形态。相比于其它四种染色方法,瑞氏-姬姆萨染色后能清晰地观察到圆形精子细胞膜和核膜的边缘,核质分明,与其它时期生精细胞区别明显。瑞-姬染色法是一种简单、有效的鉴定圆形精子形态的染色方法。
To compare five staining methods on mouse round spermatid（ROS）morphology evaluation and to find a relatively simple and effective method of identifying ROS.Testicular cell suspension was obtained from sexual maturation mouse.To compare the morphology of ROS,germ cells were stained separately by Wright＇s staining,Wright-Giemsa staining,Giemsa staining,Magenta staining and Hematoxylin staining.Compared to the other staining methods,Wright-Giemsa staining method was the best one.Wright-Giemsa staining is a convenient and effective method which can be used to identify ROS morphology.
