摘要
为了探讨亚牛磺酸的浓度对猪胚胎体外培养效果的影响,试验以孤雌激活胚胎为研究对象,将NCSU-23中的亚牛磺酸浓度从原来的5mmol/L降低到2.5,1.0,0.5mmol/L,检测猪卵母细胞激活后的分裂率和囊胚发育率。结果表明:各种浓度亚牛磺酸处理后的分裂率和囊胚发育率(30.7%、27.6%、22.8%、26.5%)差异不显著(P>0.05);但不使用亚牛磺酸则导致囊胚发育率极显著下降(30.7%vs1.4%,P<0.01)。结果说明亚牛磺酸是猪胚胎培养液必不可少的重要成分,但其浓度可下调至0.5mmol/L。
The effect of hypotaurine on the in vitro cultivation of porcine embryos was investigated in this study.Decreasing the concentration of hypotaurine in NCSU-23 medium from 5 mmol/L to 2.5,1.0 or 0.5 mmol/L did not affect the cleavage rate and blastocyst yield of activated porcine oocytes(P0.05).However,eliminating hypotaurine from NCSU-23 medium resulted in a dramatically decrease in the blastocyst development(30.7% vs 1.4%,P0.01).The result shows hypotaurine is an important component for porcine embryo culture and its concentration can be declined to 0.5 mmol/L.
