采用全胚胎培养法研究酒精对胚胎发育的毒性影响 被引量:5

摘要 目的化学物质的毒性作用常因其在体内代谢而变化。为探讨酒精在代谢条件下的发育毒性,本研究应用植入后全胚胎培养技术研究了酒精对胚胎发育的影响。方法:孕95天大鼠胚胎于体外条件下移植分离培养在含40、100、20、40mg/dl内含S9的即可离心大鼠血清培养基中连续培养48小时,观察酒精对胚胎生长发育和器官形态分化的影响。结果:40mg/dl+S9组与对照组各指标无显著性差异,100mg/dl+S9组则导致胚胎的体长、DNA、蛋白质含量及部分器官形态评分呈剂量相关的下降,但前脑、中脑评分并无影响。随染毒剂量增加,胚胎的绝大多数指标显著降低,胚胎致畸率和死亡率相应增高。结论:酒精在S9作用下转化为乙醛后,毒性作用的靶器官发生改变,表现为肢芽、心脏等器官发育异常为主,代谢产物乙醛也有一定的致畸作用。 Objective:The characterisations of chemicals substances often were changed with its metabolism.To explore the developmental toxicity of alcohol mechanism,especially,in metabolised system, this study used post-implantation whole embryo culture to observe the effect of embryo development induced by alcohol.Methods:9.5 days rat embryos were exploited and cultured in immediately centrifuged serum(ICS)that comtained S 9 and 40,100,200,and 400mg/dl alcohol for 48h.The dffect of alcohol to embryo growth development and organ morphological differentiation were observed .Results:40mg/ml+S 9 group was not significantly different from thd controls whereas the higher concentration resulted in a concentration related decrease in head length,contains of DNA,and Brown's developmental morphological score.The scores of forebrain and midbrain have not been effected,but other indexes were significantly reduced even embryolethaliyt and dysmorphogenesis high increasingly.Conclusions:Owing to the effects of metabolised system S 9,alcohol was transferred into acetaldehyde and its targets of developmental toxicity were changed. Embryolethality was increased obviously. It suggests that acetaldehyde plays some roles in teratogenesis
