摘要
5-氮杂胞嘧啶经六甲基二硅胺烷硅烷化保护,再与1-氯-3,5-对甲基苯甲酰基-脱氧-D-呋喃核糖在四氯化锡的作用下缩合得到1-(3,5-二-O-对氯苯甲酰基-2-脱氧-β-D-呋喃核糖)-5-氮杂胞嘧啶,最后经脱保护、重结晶制得地西他滨。
Decitabine was synthesized from 5-azacytosine by silanization with hexamethyldisiliz-ane,condensing with 1-chloro-3,5-di-O-p-toluoyl-2-deoxy-D-ribofuranosyl catalyzed by stannic chloride to get 1-(3,5-di-O-p-toluoyl-2-deoxy-β-D-ribofuranosyl)-5-azacytosine,followed by deprotection and recrystallization.
