摘要
以各地产西洋参为研究对象,对影响中药材随机扩增多态性DNA(RAPD)的若干问题进行探讨。研究发现,中药材DNA模板的降解程度、纯度、浓度,Tap酶的浓度,Mg2+浓度以及循环参数等诸多因素皆会不同程度的影响扩增结果,同时推荐一个较为适用的循环参数。
Random Amplified Polymorphic DNA (RAPD) method is efficient for the probation of plant species,many scientists have used this technique to identify Chinese herbs in recent years.However,it was found that some factors may affect the results of RAPD.These factors include Taq DNA polymerase concentration,Mg2+concentration,BSA,template DNA,et al.Therefore,it is important to study these factors for the credibility of RAPD.
