摘要
应用银染聚丙烯酰胺凝胶方法，对假性肥大型肌营养不良（DMD／BMD）基因5＇CA，3＇CA重复后列进行扩增片段长度多态性分析。结果表明，这两位点在中国人群中都具有长度多态性。应用这两位点对DMD／BMD家系进行了单体型连锁分析。实验表明此两位点可作为中国人的DMD／BMD基因的理想遗传标记，可对DMD／BMD家系进行基因诊断。
In this study,primers specific for two locus 5'CA, 3'CA in template DNA by polymerase chain reaction- The PCR products were separated in 6% denaturing polyacrylamidegels and displayed by silver staining. The two microsateuite 5'CA, 3'CA used in this studywere evaluated. The polymorphism information centants (PIC) of the two locus of 5'CA, 3'CA were 0. 64,0. 32 respectively. The two Locus are useful in genetic linkage analysis TwoDMD/BMD families were analyzed by Amp-FLP Linkage analysis.
