摘要
目的对比人脑胶质瘤细胞的分离纯化方法,并观察其形态学特征。方法取术后人脑胶质瘤新鲜组织,采用冷胰酶消化法和机械分离法分离后结合速差贴壁法纯化处理进行原代培养。采用吉姆萨染色对培养细胞进行形态学观察。结果在细胞产量方面冷胰酶消化法优于机械分离法。结论采用冷胰酶消化法,可以显著降低胰酶对人脑胶质瘤细胞的损害,是可靠的人脑胶质瘤细胞的分离纯化方法。
Objective To compare two methods of the human brain glioma cells isolated from the patients, and to observe the morphological characteristics. Methods The human glioma cells were isolated from glioma tissue of patients by the cold- trypsin digestion and the mechanical separation, and were purified and cultured with differential attachment treatment. The cultured cells were observed by Giemsa staining. Results The cold-trypsin digestion method was superior to the mechanical separation in the aspect of cell yield. Conclusion The cold-trypsin digestion can significantly reduce the damage of trypsin to the human brain glioma cell. It is a reliable method of human brain glioma cell separation and purification.
