摘要
达乌尔鼠兔肝细胞线粒体DNA(mtDNA),经限制性内切酶HindⅡ、HindⅢ、EcoRⅠ和BamHⅠ酶切后分别产生 5、4、3、2个片段。通过琼脂糖凝胶电泳对这些片段进行测定,并画出其酶切图谱。高原鼠兔肝细胞mtDNA经限制性内切酶EcoRⅠ和 BamHⅠ酶切后,分别产生4、2个片段,对其片段的分子量也进行了测定。测定结果,达乌尔鼠兔肝细胞mtDNA的分子量为10.25MD(兆道尔顿),大小为16.25kbp(千碱基对);高原鼠兔肝细胞mtDNA的分子量为9.31MD,大小为15.066kbp。并对两种鼠兔的限制性内切酶片段进行了比较讨论。
mtDNA from Ochotona daurica liver was deaved by HindⅡ , HindⅢ , EcoRl and BamHI into 5, 4, 3, and 2 fragments. The sizes and orders of these fragments in the respective genomes were determined with data obtained from agarose gel el ectrophores is. mtDNA from O.curzoniae Hver was cleaved by EcoRI and BamHI into 4 and 2 fragm-ents. The sizes were determined by the same way as in O.daurica. The molecular wei-ght of O. daurica mtDNA was found to be 10.25 MD and about 16.59 kbpt the molec-ular weight of O.curzoniae mtDNA was 9.31 MD and about 15.066 kbp.The differences of restrictive fragments between mtDNA from O,daurica and O.curzoniae were discussed.
