摘要
全长cDNA文库是目前基因鉴定、基因启动子区确定和功能基因组研究的必要工具,为快速、高效获得完整的基因全序列信息提供了有效途径。高质量的全长cDNA文库要求较高比例的大片段全长cDNA克隆。Vector-Capping正是一种简单而有效的构建高质量全长cDNA文库的方法。在此重点阐述Vector-Capping法的建库依据、技术流程、特点和应用进展。
Nowadays full-length cDNA Library plays an essential role in identifying genes,determining their promoter regions and studying the functional genomics.It provides an effective way to obtain the complete gene information rapidly.One of the requirements for a high-quality cDNA library is the high ratio of long-sized full-length cDNA clones.The Vector-Capping method is exactly a simple method for constructing a high-quality full-length cDNA library.In this paper,the theory and protocol of this method was described in detail,and its characteristics and application was also mentioned.
