马鼻疽弱毒菌苗对豚鼠的免疫试验

摘要 将马鼻疽强毒菌C67-40株经60Co-γ射线照射和在温度递增的肉汤中传代培养,使其毒力逐渐减弱。通过反复试验筛选,培育出C67-4060Co(7)兼高温178代、C67-40高温180代两株弱毒菌株。根据菌苗菌株的要求,将其分别给雄豚鼠(350 g左右)腹腔接种0.2-6.2亿菌体和0.2-1.4亿菌体,结果100%雄豚鼠不发病,60Co(7)兼高温178代致弱菌株的安全性比强毒菌株提高62倍;以其0.2亿菌体免疫豚鼠并以0.3亿强毒菌攻击,获得85.7%-87.5%的保护率;将其通过培养基37℃传代培养30代和豚鼠6代。其安全性和免疫原性与原菌株比较差异不大,说明该两株鼻疽弱毒菌株毒力稳定。 The virulent C67-40 strain of Pseudomonas mallei was gradually attenuated by repeated irradiation with 60Co-γ ray and/or serial subcultivation in broth at gradient temperature(38. 5 -45℃). By screening test two variants, 60C67-40·60Co (7) -HT (178) and C67-40·HT(180), were selected as seed cultures in order to prepare the attenuated vaccines for guinea pigs. It was shown that most of them immunized with these vaccines at dosags of twenty million bacteria have become resistant to challenge of large dose (thirty million) of C67-40 strain, the protective rate was 85.7%-87.5%., No significant changes in their virulence and immu-nogenicity were found after successive subcultivation and guinea pig inoculation.
