大肠癌微血管密度和血管内皮生长因子表达的研究 被引量:10

STUDY ON THE MICROVESSEL DENSITY AND EXPRESSION OF VASCULAR ENDOTHELIAL GROWTH FACTOR IN HUMAN COLORECTAL CANCER

摘要 目的探讨大肠癌间质微血管密度(MVD)和血管内皮生长因子(VEGF)表达与肿瘤浸润和转移的关系。方法应用CD34抗体和VEGF抗体,采用免疫组化S-P法对56例手术切除的大肠癌患者进行血管标记和染色,并取10例正常组织对照。结果有淋巴结转移组大肠癌MVD、VEGF表达强度与无淋巴结转移组、正常对照组组间比较,均有显著性差异(P<0.01),且MVD与VEGF表达两者呈正相关(r=0.92)。结论大肠癌间质内微血管密度和VEGF表达强度与淋巴结转移密切相关,两者均可作为预测大肠癌发生转移的指标表1正常对照组、非转移组、转移组MVD和VEGF的表达(x±s)类型nMVDVEGF正常对照组1068.20±13.691.00±0.94非转移组3674.75±14.041.33±0.93转移组2095.65±18.902.45±0.692.2MVD与组织学分型的关系统计学处理,显示微血管计数在不同组织学分型的大肠癌中无显著性差异(P>0.05)(见表2)。表2MVD和VEGF的表达与组织学类型的关系(x±s)组织学类型nMVDVEGF高分化腺癌1781.71±19.161.76±1.15中分化腺癌2081.05±15? Objective To study the relationship between the microvessel density (MVD) and expression of vascular endothelial growth factor (VEGF) and lymph node metastasis of human colorectal cancer. Methods Immunohistochemical analyses (S P method) using antibodies against CD 34 and VEGF were carried out on archival specimens of 56 human colorectal carcinomas and 10 normal controls. Results Microvessels were quantitated by light microscopy (200×),and the intensity of staining for VEGF (400×) was assessed on a scale of 0~3+. Microvessel density were greater in metastatic tumors than in nonmetastic tumors and normal colon tissue ( P <0.01). Expression of VEGF was higher in metastatic than nonmetastatic carcinomas ( P <0.01). They had a strong correlation ( r =0.92). Conclusions Microvessel density and VEGF expression were helpful in predicting patients at risk for metastasis from colorectal cancer.
