摘要
目的:研究含重复序列(GAA)n·(TTC)n的菌体在传代培养中氨苄青霉素对其遗传突变的影响。方法:采用分子克隆、PCR等分子生物学实验技术。结果:在100mLLB液体培养基中加入100mg/mL氨苄青霉素35μL对重复序列有明显删减。结论:治疗与重复序列相关的神经系统遗传疾病可以通过促进重复序列的有效删减,将重复序列数目控制在阈值之内来完成。
Objective:To explore the impact of ampicillin on genetic mutations of(GAA) n·(TTC) n repeat sequence bacteria in the passage culture.Methods:Molecular cloning,PCR and other molecular biological techniques were used in the study.Results:100mg/ml ampicillin35μl added to 100ml LB liquid medium remarkably deleted the repeat sequences.Conclusion:Treatment of genetic diseases of nervous system associated with repeat sequence can be achieved by promoting the effective deletion of repeats and keeping the number of repeats within the control threshold.
