失血性休克大鼠血清TXB_2等内源性介质水平变化及意义

Serum levels of TXB_2, 6-keto -PGF_1α and 8-iso -PGF_2α of hemorrhagic shock rats, and their significances

摘要 目的检测失血性休克大鼠血清血栓素B2(TXB2)、6-酮-前列腺素F1α(6-keto-PGF1α)及8-异前列腺素F2α(8-iso-PGF2α)水平探讨其与失血性休克的不同程度及不同时相的相关性。方法雄性Wistar大鼠50只,随机分为5组:正常对照组(sham hemorrhage shock,SHS)10只;重度失血性休克(refractory hemorrhagic shock,RHS)组10只;中度失血性休克组(moderate hemorrhagic shock,MHS)30只,按休克后采血时间的不同又分为休克30 min组(MHS-30 min)10只、休克60 min组(MHS-60 min)10只及休克120 min组(MHS-120 min)10只。采用ELISA方法及酶法分别对各组血清TXB2、6-keto-PGF1α及8-iso-PGF2α水平进行检测。结果 (1)各组TXB2、6-keto-PGF1α及8-iso-PGF2α水平比较:①MHS组及RHS组TXB2、6-keto-PGF1α及8-iso-PGF2α水平均显著高于SHS组(P均<0.01);②RHS组与MHS组比较,TXB2及8-iso-PGF2α水平显著升高(P<0.01),6-keto-PGF1α水平显著降低(P<0.05);③MHS三不同时相组之间比较:TXB2水平呈上升-下降-上升的趋势,三组间两两比较,TXB2水平均具有显著性差异(P<0.01),MHS-120 min组水平最高;6-keto-PGF1α水平呈上升-上升-下降的趋势,三组间两两比较,6-keto-PGF1α水平均具有显著性差异(P<0.01),MHS-60 min组水平最高;8-iso-PGF2α水平随休克时间的延长呈持续上升的趋势,三组间两两比较,8-iso-PGF2α水平均具有显著性差异(P<0.01);(2)各组TXB2/6-keto-PGF1α(T/K)比值比较:①MHS各组及RHS组T/K比值显著高于SHS组(P均<0.01);②RHS组与MHS组比较,T/K比值显著升高(P<0.01);③MHS三不同时相组之间比较:T/K比值呈上升-下降-上升的趋势(三组水平均高于SHS组),三组间两两比较,T/K比值均具有显著性差异(P<0.01),MHS-120 min组T/K比值最高。结论不同程度失血性休克大鼠血清TXB2、6-keto-PGF1α、8-iso-PGF2α水平及T/K比值存在显著性差异,且随着休克的发展呈动态变化,因此监测血清TXB2、6-keto-PGF1α及8-iso-PGF2α水平对了解失血性休克条件下机体的炎症反应具有重要意义。 Objective To detect the serum levels of thromboxane B2(TXB2),6-keto-prostaglandin Flα(6-keto-PGF1α) and 8-iso-prostaglandin F2α(8-iso-PGF2α) of hemorrhagic shock(HS) rats,so as to study the correlations between these items and variation of different degree and phase in HS.Method Fifty male Wistar rats were randomly divided into 5 groups(n=10): Sham hemorrhagic shock(SHS);refractory hemorrhagic shock(RHS);moderate hemorrhagic shock(MHS) which were subdivided into MSH-30 min,MSH-60 min and MSH-120 min following the time after shock.The serum levels of TXB2,6-keto-PGF1α and 8-iso-PGF2α were detected respectively.Results 1) Compared with SHS,the levels of TXB2,8-iso-PGF2α and the ratio of TXB2/6-keto-PGF1α(T/K) increased in MHS and RHS group(P0.01),and the severer of HS was,the more the items increased.The levels of 6-Keto-PGF1α decreased in MHS and RHS group(P0.01),and the severer of HS was,the more the items decreased.2) Compared among three groups of MHS-30,MHS-60 and MHS-120,the levels of TXB2,8-iso-PGF2α and the ratio of T/K decreased first,then increased in MHS-120(P0.01).The levels of 6-Keto-PGF1α increased first,then decreased in MHS-120(P0.01).Conclusion The serum levels of TXB2,6-keto-PGF1α and 8-iso-PGF2α are different in rats subjected to HS of different degree and vary dynamics with the prolongation shock time,which can be helpful to detect the levels of TXB2,6-keto-PGF1α and 8-iso-PGF2α to understand the variation of inflammation in rat subjected to HS.
