摘要
目的探讨抗菌肽tachyplesin-I对胃腺癌BGC-823细胞膜的作用机制。方法 BGC-823细胞用不同质量浓度tachy-plesin-I处理,再用台盼蓝拒染法测定活细胞率,荧光分光光度计检测细胞膜荧光素渗漏,激光共聚焦显微镜和电镜观察细胞膜的破坏情况。结果低质量浓度(≤25mg/L)tachyplesin-I对细胞存活没有明显影响,细胞膜荧光素渗漏较少[(15.14±0.05)~(17.84±0.68)];高质量浓度tachyplesin-I(≥50mg/L)使细胞存活率明显下降,细胞膜荧光素渗漏较多[(60.54±4.08)~(67.57±0.18)],并进一步引起细胞死亡。结论 tachyplesin-I对BGC-823细胞膜存在质量浓度阈值,低质量浓度tachyplesin-I引起膜可逆性通透,高质量浓度tachyplesin-I造成细胞膜严重损伤。
Objective To investigate the mechanism of action of tachyplesin-I on gastric adenocarcinoma BGC-823 cell membrane. Methods After treatment with different concentrations of tachyplesin-I, the viability, membrane fluorescein leakage and destruction of BGC-823 cells were detected by trypan blue excluding test, spectrophotofluorometer, confocal microscopy and electron microscopy, respectively. Results Low concentrations (≤25 mg/L) of tachyplesin-I resulted in intact viability and a little of fluorescein leakage [(15.14±0.05)~(17.84±0.68)] in BGC-823 cells. However, high concentrations (≥50 mg/L) of tachyplesin-I induced lower viability, a great deal of fluorescein leakage [(60.54±4.08)~(67.57±0.18)], and progressive death. Conclusion There exists a concentration threshold of tachyplesin-I on BGC-823 cell membrane. Low and high concentrations of tachyplesin-I can lead to the reversible permeation and severe damage of cell membrane, respectively.
