摘要
目的:对HLA-A2限制性细胞毒性T淋巴细胞(cytotoxic T lymphocyte,CTL)表位HPV18E77-15进行氨基酸置换修饰,探讨修饰肽的免疫原性。方法:采用细胞毒实验(51Cr释放法),以包被了九肽的T2细胞作为靶细胞,以九肽诱导的HLA-A2阳性人外周血单个核细胞为效应细胞,观察目标肽是否具有诱导特异性CTL杀伤效应。结果:修饰肽符合HLA-A2分子限制性细胞毒性T细胞的表位要求,具有特异性细胞毒性T细胞诱导活性。结论:修饰肽具有较强的抗原性,可以作为高危型HPV感染治疗性肽疫苗分子设计的候选表位。
Objective:To investigate the immunogenicity of peptide modified by amino acid substitution for the cytotoxic T lymphocytes(CTLs) epitope HPV18E77-15 restricted by HLA-A2. Methods:51Cr release assay was used to determine their abilities of inducing the generation of specific CTLs. Results:The modified peptide TLQDIVLHV met the requirements of HLA-A2-restricted CTL epitopes and had the abilitiy of inducing the generation of specific CTLs. Conclusion:The modified peptide TLQDIVLHV may have better antigenicity and affinity to HLA-A2,and can be used as one of the HLA-A2 restricted candidate epitopes for therapeutic peptide vaccine against HPV infection.
