摘要 目的 pEGFP-N1/BMP-2真核表达质粒转染骨髓间充质干细胞,并检测其在靶细胞中的表达。方法脂质体将pEGFP-N1/BMP-2真核表达质粒转染入BMSCs,荧光显微镜下观察绿色荧光,计算转染率,利用ELLISA法检测目的基因在靶细胞中的表达,免疫组化染色检测成骨特异性生化指标(I型胶原、ALP及骨钙素)。结果本实验成功的利用脂质体将质粒转染入BMSCs,目的基因可以持续高表达,并促进细胞向成骨方向分化。结论为进一步利用BMP-2基因修饰组强Ⅰ程骨促进成骨提供实验基础。 Objective The eukaryotic expression vector carrying BMP-2 gene was transfected into BMSCs by liposome,and detected the expression.Methods The eukaryotic expression vector carrying BMP-2 gene was transfected into BMSCs by liposome.fluorescence microscope observe cells culture after BMSCs transfected.The protein of cells transfected was detected by ELISA and immunohistochemical method.Results The plasmid was transfected into BMSCs,the gene kept on expressing.Conclusion It provies the base of engineering bone modified by BMP-2 gere.
