摘要 目的 探讨亚甲蓝光化学法血浆病毒灭活对血浆有效成分的影响.方法 使用血凝仪对血浆进行Ⅷ活性和纤维蛋白原含量检测 使用分光光度计测定血浆总蛋白浓度的变化.结果 新鲜冰冻血浆经MB-P病毒灭活后血浆Ⅷ因子活性为(0.66±0.06)IU/ml,比灭活前(0.75±0.07)IU/ml降低,纤维蛋白原含量为(1.31±0.1)mg/ml,比灭活前(2.19±0.2)mg/ml减少,两者比较差异均有显著性(P<0.05),总蛋白含量比较差异无显著性(P>0.05).结论 在制备冷沉淀时,可考虑先将新鲜冰冻血浆制备冷沉淀后再将血浆进行病毒灭活.灭活后血浆对于临床需要补充血浆蛋白的患者影响不大 对于需要输注Ⅷ因子、纤维蛋白原的患者则会有一定影响. Objective To analyze the effect of methylene blue (MB) photochemical method for virus inactivation on the active constituent of plasma. Metbods Detection was made on the plasma total protein concentration by spectrophotometer, fibrin content and blood coagulation factor by coagulometer,before and after methylene blue photochemical viral inactivation. Results After virus was inactivated through methylene blue method, plasma proteins were not affected significantly(P〉0.05 ), activities of the factor Ⅷ were decreased from ( 0.75 ± 0.07 ) IU/mlto ( 0.66 ± 0.06 ) IU/ml, fibrin content were decreased from ( 1.31 ± 0.1 ) mg/ml to ( 2.19 ± 0.2 ) mg/ml, activities of the factor Ⅷ and fibrin content were significantly decreased ( P〈 0.05 ). Conclusion The preparation of cryoprecitation should be done before the virus inactivation of the fresh frozen plasma. There is no great effect on the patients who need replenish plasma protein, meanwhile, there is certain effect on the patients who need transfuse the factor Ⅷ and fibrin, when the plasma was treated by virus inactivation.
