内皮素-1对膀胱癌环氧化酶-2表达的影响及意义

Effect and clinic significance of endothelin-1 on cyclooxygenase-2 expression in bladder cancer

目的研究内皮素-1(ET-1)对膀胱癌细胞株T24环氧化酶-2(COX-2)表达的影响及意义。方法应用不同浓度(0μg/L、10μg/L、100μg/L)的ET-1刺激T24细胞6 h后,用RT-PCR和Western-blot检测其COX-2 mRNA和蛋白水平变化。结果 ET-1可使T24细胞COX-2的mRNA和蛋白表达明显升高,与对照组比较有显著性差异(P均<0.05)。结论 ET-1可使T24细胞COX-2的表达上调,这种调节可能在膀胱癌的复发和进展中发挥重要作用。

Objective It is to observe the influence and clinic significance of Endothelin-1 on the cyclooxygenase-2（COX-2） expression in bladder cancer T24 cell line.Methods The expression of COX-2 in the T24 cell was detected by RT-PCR and Western blot assay after they were stimulated with Endothelin-1（0μg/L,10μg/L,100μg/L） for 6h.Results The COX-2 expression in T24 cell was significantly increased after exposure to ET-1 for 6 h（P〈0.05）.Conclusion Endothelin-1 can induce upregulation of COX-2 in T24 cell and the regulation maybe play an important role in the recurrence and development of bladder cancer.
