摘要
利用高压匀浆法破碎、硫酸铵分级盐析、DEAE-Sephadex A-50阴离子交换层析以及Sephadex G-150凝胶过滤层析对废弃啤酒酵母细胞线粒体中乙醛脱氢酶(ALDH)进行了提取研究。结果表明,经过纯化后的ALDH酶液,纯化倍数为29.6,产率为6.7%。酶的亚基分子量经检测为55kDa。
Mitochondrial aldehyde dehydrogenase （ALDH） was extracted fi＇om yeast residue by using high-pressure homogenization, ammonium sulfate fractionation, ion-exchange chromatography on DEAE-Sephadex A-50 and gel filtration chromatography on Sephadex G-150. The results showed that fold of purification of ALDH was 29.6 and the yield was 6.7%. Molecular weight of subunit was 55kDa.
