Tadenan对早期糖尿病大鼠膀胱组织中NGF及P物质表达的影响

摘要 目的探讨Tadenan对早期糖尿病大鼠膀胱组织中神经生长因子(NGF)及P物质表达的影响。方法将36只雄性Wistar大鼠随机分为对照组(10只)、糖尿病组(10只)、治疗组(10只)、药物组(6只)。糖尿病组和治疗组大鼠先制作成糖尿病模型。饲养4周后,治疗组及药物组大鼠给予Tadenan 100 mg/(kg.d),溶于2 ml花生油中经胃管注入;对照组及糖尿病组大鼠给予同等剂量花生油赋形剂。以上动物共饲养8周。8周后处死,完整切取膀胱组织。采用RT-PCR及免疫组化法、ELISA法检测膀胱组织中NGF、P物质。结果糖尿病组NGF、P物质明显降低(P均<0.05);与糖尿病组相比,治疗组NGF及P物质显著升高(P<0.05)。结论 Tadenan可增加膀胱组织中NGF及P物质表达,推测这是Tadenan对糖尿病膀胱具有保护作用的机制。 Objective To investigate the effects of Tadenan on nerve growth factor(NGF) and substance P in the bladder 4 weeks after diabetes induction. Methods 36 Wistar rats were divided into four groups: control group( n = 10), diabetic group( n = 10), treated group( n = 10, diabetic rats were fed with Tadenan) , drug group( n = 6 ,normal rats were fed with Tadcnan). Animals in diabetic group and treated group were successfully induced into diabetic model. 4 weeks later, treated group and drug group were given Tadenan 100 rag/( kg ~ d), control group and diabetic group were given the same dose of oil for control. All the rats were fed 8 weeks. 8 weeks later, the bladders were dissected. Its were used by RT-PCR and ELISA to evaluate the expression of NGF, and used immunohistoehemistry and ELISA to detect the expression of substance P. Results Compared with the diabetic group, the expression of NGF and substance P were greatly increased in the treated group( P 〈 0.05). Conclusions Tadenan increase the express of NGF and substance P, that is suspected is mechanism of Tadenan protecting.
