摘要
目的:探讨人血T细胞在异种肾移植(猪→人)间接体内模型中的变化。方法:以模拟人体生理条件下的体外灌注系统,将人新鲜血灌注离体猪肾25只(实验组),人肾12只(对照组);利用流式细胞术,检测灌注前后不同时间内人血T细胞的变化。结果:经猪肾灌注的人血T细胞在灌注一开始即锐减,由灌注前的678±34下降至324±21,下降了约52%;随着灌注时间的延长,其减少的幅度逐渐减弱。而对照组T细胞在灌注前后无明显变化。结论:与同种移植不同,异种器官接通血流一开始,人血T细胞锐减,表明T细胞可能通过与猪肾血管内皮细胞的粘附,参与了异种移植排斥反应。
OBJECTIVE To investigate the changes of T cell and in an ex vivo study of pig to man renal xenotransplantation. METHODOLOGY Twenty five pig kidneys and 12 human kidneys(as control)were perfused with human fresh blood in an ex vivo model of pig to man renal xenotransplantation.T cells labeled with immunofluorescent McAbs were analyzed by flow cytometer before and after perfusion. RESULTS In the first minute of xenogenic perfusion,human T cells decreased 52%(from 67 8±3 4 to 32 4±2 1),and the magnitude of T cell decrease lessened there after.No significant change was observed in control group before and after perfusion. CONCLUSION In this study,it is found that human T cells may play a role in the cell mediated xenogeneic rejection of pig to man renal xenotransplantation possibly by adhering to procine renal endothelial cells during the perfusion.
