摘要 [目的]探讨HIF-1α、Angiopoietin-2(Ang-2)在原发性肝细胞癌(HCC)中的表达及意义.[方法]检测43例HCC中HIF-1α、Ang-2的表达情况,探讨其与肝癌血管生成等临床病理因素的关系.[结果]在43例肝癌标本中,HIF-1α、Ang-2蛋白阳性表达分别为:60.5%、79.1%.而7例正常的肝组织阳性表达分别为:14.2%、14.2%,两者相比差异具有显著性( P 〈0.05).HIF-1α表达与肿瘤的大小、分化程度有关, Ang-2表达与肿瘤的大小、有无门静脉、胆道癌栓有关.HIF-1α、Ang-2阳性表达呈正相关性.[结论]①HIF-1α在HCC中表达强于正常组织,且与肿瘤的大小、分化程度有关;而与HBsAg、门静脉癌栓无明显相关性.②Ang-2仅在HCC中表达.其表达与分化程度、门静脉癌栓有关;而与肿瘤的大小、HBsAg检测结果无明显相关性.③HCC组织存在着HIF-1α, Ang-2蛋白的表达上调并与MVD相关;三者协同作用,调节HCC的血管新生,共同促进HCC的发展. [Objective]To explore the expression of HIF-la and angiopoietin-2(Ang-2) in primary hepatocellular carcinoma(HCC) and its significance. [Methods]The expression of HIF-la and Ang-2 were detected in 43 cases of HCC. The relationship between the expression and clinicopathological factors of the angiogenesis of HCC. [Results]The positive expression rate of HIF-1α and Ang-2 proteins in HCC tissues were 60.5% and 79.1%, and their positive expression rates in normal hepatic tissues were 14. 2% and 14.2%, respectively. There was significant difference in the expression between HCC and normal hepatic tissues( P 〈0.05). HIF- 1α was correlated with the tumor size and differentiation extent, and Ang-2 was correlated with the tumor size, the existence of portal vein and cancer embolus in the biliary tract. The positive expression of HIF-1α was correlated directly with the expression of Ang-2. [Conclusion]The expression of HIF-1α in HCC is higher than that in normal tissues and correlated with the tumor size and differentiation extent, but not significantly correlated with HBsAg and cancer embolus of portal vein. Ang-2 is expressed only in HCC and its expression is correlated with differentiation extent and cancer embolus of portal vein, but not significantly correlated with the tumor size and HBsAg. The expression of HIF-1α and Ang 2 proteins increase in HCC tissues and are correlated with microvessel density(MVD). The synergistic effect exists among them, and regulates the angiogenesis of HCC, and promotes the development of HCC.
