摘要
目的:探讨苦参碱对博来霉素(BLM)诱导的大鼠肺纤维化模型肺组织病理变化的影响。方法:120只雄性SD大鼠采用随机数字表法分为正常对照组(A组)、模型对照组(B组)、泼尼松组(C组)、苦参碱50 mg组(D组)、苦参碱200 mg组(E组),使用博来霉素(5 mg/kg)气管注射法复制肺纤维化大鼠模型,苦参碱50 mg组和苦参碱200 mg组分别经胃管注入苦参碱50、200 mg/kg,1次/d;泼尼松组经胃管注入醋酸泼尼松0.56 mg/Kg,1次/d。分别于7、14、28 d每组各随机选取大鼠8只,处死,称取肺重及体重后取右肺组织行苏木精-依红(HE)、Masson胶原染色,并计算肺泡炎及肺纤维化积分。结果:模型组肺泡炎和肺纤维化积分显著高于正常对照组(P<0.01);苦参碱50 mg组、苦参碱200 mg组及泼尼松组肺泡炎和纤维化积分显著低于模型对照组(P<0.01);而苦参碱200 mg组积分低于苦参碱50 mg组及泼尼松组(P<0.01)。结论:苦参碱能减轻博莱霉素诱导的大鼠肺纤维化模型的肺泡炎和肺纤维化程度,并具有剂量依赖性。
Objective:Study effect of pathological change treated with matrine in the pulmonary fibrosis rats induced by bleomycin.Methods:120 adult male SD rats were randomly divided into control group(group A),Bleomycin group(group B),Prednisone Acetate group(group C),Matrine50mg group(group D) and Matrine200mg group(group E).Pulmonary fibrosis model was established by injected with Bleomycin by intratracheal.Every rats was raised drugs through a gastric tube one times a day :0.56 mg/kg Prednisone Acetate in Group C,50 mg/kg matrine in Group D and 200 mg/kg matrine in Group D.At 7、14、28 d,eight rats was killed in every groups.Weighted the weight of bodies ang lungs.After HE and Masson staining,right lung tissue was calculated the points of alveolitis and pulmonary fibrosis.Results:The points of alveolitis and pulmonary fibrosis is highest in group B,and lowest in group A(P〈0.01).In group E,it is lower than in groupB,C and D(P〈0.01).Conclusions:Matrine reduce alveolitis and pulmonary fibrosis in rats induced by bleomycin.And the effect has dose-dependant.
