摘要
目的:通过双向电泳技术分离骨骼肌蛋白质组份,探讨悬吊后肌萎缩组股直肌与正常骨骼肌的差异蛋白。方法:建立大鼠尾悬吊模型,使大鼠后肢去负荷,两周后取大鼠股直肌进行双向电泳技术分离,使用PDQuest软件对凝胶图像进行差异蛋白组学分析。结果:研究发现在悬吊所致肌萎缩后有23个点发生了明显而稳定的改变,其中下调的蛋白有18个,上调的有5个。结论:悬吊致肌萎缩后,股直肌收缩蛋白、骨架蛋白、运输蛋白含量下调,而与代谢有关的蛋白或酶的表达有升有降,表现了在肌萎缩过程中代谢调节的复杂性。
Objective: To identify major muscle proteins responding significantly to the unloading,Compared with unsuspended controls,we examined protein expression in the rat muscle during 2-week hindlimb suspension by 2-DE.Methods: Establish the model of hindlimb suspension,to make rat unloading,then examined the proteins by 2-DE after 2-week.Result: we found 23 different expression proteins during unloading.Among these,we observed 18 decreased,and 5 increased.Conclusions: After 2-week hindlimb suspension,These results of unloading of skeletal muscle has a decrease change overall contractile proteins,cytoskeleton proteins, transport proteins,but the metabolic proteins has both increase and decrease,it implies that the metabolic regulation is a complex process.
