苏木水提物对皮肤移植小鼠CD4^+ CD25^+ T细胞及IL-10 TGF-β1的影响 被引量:1

Effect of watery extract of caesalpinia sappan L on CD4^+ CD25^+ T cells and IL-10,TGF-β1 in allografted mice

摘要 目的探讨苏木水提物在小鼠同种皮肤移植中对CD4+ CD25+ T细胞及IL-10、TGF-β1水平的影响。方法 C57BL/6和BALB/c小鼠分别为供体和受体,建立皮肤移植模型。实验分为对照组、苏木组、苏木加半量环孢素A(CsA)组、CsA组。术前3 d给药,术后逐日观察移植皮片存活情况;术后3 d、7 d时检测脾组织中CD4+ CD25+ T细胞数量变化及血清中IL-10、TGF-β1表达情况。结果与对照组比,苏木组、苏木加半量CsA组、CsA组移植皮片存活时间明显延长(P<0.01),移植后3 d、7 d时,CD4+ CD25+ T细胞数量增加(P<0.01),IL-10、TGF-β1水平在移植后7 d时均不同程度上调(P<0.05)。结论苏木水提物可延长同种移植物的存活时间,上调CD4+ CD25+ T细胞及IL-10、TGF-β1的水平,有较强的免疫抑制作用。 Objective To investigate the effect of Caesalpinia Sappan L on CD4+ CD25+ T cells and IL-10,TGF-β1 after allo-transplantation in mice.Methods Skin of C57BL/6 mice were transplanted to BALB/c mice.Groups include control group,Caesalpinia Sappan L group,CsA group and Caesalpinia Sappan L plus semidose CsA group.Animal were given above drugs three days before operation.Survival conditions of grafted skin were observed daily;Numbers of CD4+ CD25+ T cells in sleep were examined by flow cytometry;The levels of IL-10、TGF-β1 in peripheral blood were tested by ELISA.Results Compared with group Control,allograft survival time of group Caesalpinia Sappan L,Caesalpinia Sappan L plus semidose CsA and CsA prolonged significantly(P〈0.01),CD4+ CD25+ T cells increased obviously on the third and seven day after transplantation(P〈0.01);and the levels of IL-10、TGF-β1 increased on the seven day after transplantation(P〈0.05).Conclusion Caesalpinia Sappan L can prolong graft survival time and up-regulate the levels of CD4+ CD25+ T cells,IL-10,TGF-β1.So Caesalpinia Sappan L has a marked immunosuppressive effect.
