摘要
利用DCR培养基,采用正交设计,诱导云南松(Pinus yunnanensis)成熟合子胚的愈伤组织,并根据实验进展,进一步精确激素浓度配比。结果表明,用5%的次氯酸钠溶液消毒,在DCR培养基中添加10 mg/L 2,4-D与1.5 mg/L 6-BA为最佳诱导激素浓度配比,用此方法较可以高效地诱导出云南松成熟合子胚的胚性愈伤组织。
Experiment was conducted on induction of embryogenic callus from mature Pinus yunnanensis zygotic embryo on DCR as minimal medium with orthogonal design.Hormone concentrations were tested.The result showed that 5% sodium hypochlorite solution and 10mg/L of 2,4-D,1.5 mg/L of 6-BA was the best condition for induction.The experiment concluded that with conditions above-mentioned,embryogenic callus could be induced with high efficiency and simplicity.
