生骨胶囊对兔骨折愈合过程中转化生长因子-β_1的影响

摘要 目的观察生骨胶囊对兔骨折愈合过程中转化生长因子-β1(TGF-β1)的影响,以进一步探讨其促进骨折愈合的机制。方法将60只健康雄性家兔随机分为治疗组和对照组各30只,在兔桡骨处用手术方法制备骨折模型,造模后即日治疗组予生骨胶囊150mg/(kg.d),对照组予骨折挫伤胶囊150mg/(kg.d)。于骨折后3d和1、2、3、4、6周每组分别抽取5只兔处死,取标本,采用免疫组化法,并利用图像分析的方法测定TGF-β1表达强度。结果骨折早期TGF-β1染色强度均值低,以后逐渐升高,成骨活跃时最高,随着修复逐渐完成,TGF-β1染色强度均值逐渐降低。骨折后3d,相当于骨折损伤反应期,对照组TGF-β1染色强度均值较治疗组高(P<0.05);治疗组在骨折后2周出现染色高峰,较对照组提前1周,比较差异有统计学意义(P<0.01)。结论生骨胶囊可以调节骨折愈合过程中TGF-β1的合成和分泌,从而促进骨折愈合,TGF-β1可能是生骨胶囊对骨折愈合过程中的一个作用环节。 Objective To observe the effect of Shenggu capsule on TGF-β1 in rabbit with fracture healing,and explore the mechanism of Shenggu capsule in improving fracture healing.Methods 60 healthy rabbits were randomly divided into two groups.Fracture rabbit model were established by operation of radius.Rabbits in treatment group were treated by Shenggu capsule,150 mg/(kg.d).Rabbits in control group were treated by Guzhe Cuoshang capsule,150 mg/(kg.d).Expression intensity of TGF-β1 were measured by immunohistochemistry and image analysis 3 day,1,2,3,4 and 6 weeks after fracture.Results Dyeing intensity of TGF-β1 was lower early fracture.The intensity of TGF-β1 was gradually increased,maximum of TGF-β1 was in active osteogenic period.The intensity of TGF-β1 in control group was higher than that in treatment group at 3 days after fracture(P〈0.05).Maximum of TGF-β1 in treatment group was in 2 weeks after fracture.It was a week earlier than the control group(P 0.01).Conclusion Shenggu capsule can regulate synthesis and secretion of TGF-β1,and improve fracture healing.TGF-β1 may be part of role in fracture healing of Shenggu capsule.
