摘要
神经鞘脂(sphingolipid)是生物膜脂质类的一个家族,它在调节脂质双分子层的流动性和亚结构域方面发挥着重要作用。神经酰胺(ceramide)作为神经鞘脂代谢的核心分子起着第二信使的作用,可介导细胞凋亡、分化、生长抑制等多种生物效应。细胞内神经酰胺浓度降低和恶性肿瘤抗凋亡机制之间的联系越来越密切,因此增加内源性神经酰胺浓度或外源性类似物(模拟神经酰胺活性)可以逆转肿瘤的耐药性,并为恶性肿瘤治疗提供全新方法 。
Sphingolipids are a family of membrane lipids with important structural roles in the regulation of the fluidity and subdomain structure of the lipid bilayer.As a central molecule in sphingolipid metabolism,ceramide induces many biological effects including cell apoptosis,differerentiation and growth inhibition.Attenuation of ceramide levels is increasingly implicated in anti-apoptotic mechanism of malignant tumour,so inducing endogenous ceramide accumulation or mimicking its actions by exogenous ceramide analogue could present a novel therapeutic modality for the treatment of malignant tumour including the reversal of drug resistance.
