B7-H4在原发性卵巢上皮性癌中的表达及临床病理意义

摘要 目的探讨B7-H4在原发性卵巢上皮性癌(EOC)中的表达情况及其临床病理意义。方法用逆转录聚合酶链反应(RT-PCR)技术及免疫组织化学PV-9000二步法研究86例原发性EOC(EOC组)、40例良性卵巢肿瘤(良性组)和10例正常卵巢组织(对照组)B7-H4 mRNA及B7-H4蛋白表达情况,并结合临床病理参数进行分析。结果 B7-H4 mRNA在3组卵巢组织中均有表达,且3组卵巢组织的 mRNA光密度值比较差异均有统计学意义(P<0.05);B7-H4蛋白在EOC组中呈细胞质和(或)细胞膜表达,阳性率为77.9%(67/86),显著高于良性组5.0%(2/40)及对照组0%(0/10)的表达(P<0.05)。EOC组的卵巢癌组织中B7-H4蛋白表达与组织病理学类型、临床分期、病理分级、患者年龄、腹水细胞学和淋巴结转移差异均无统计学意义(P>0.05)。结论 B7-H4在原发性EOC的高表达提示其可能与肿瘤的发生有关,可为卵巢恶性肿瘤的诊断及治疗提供新的依据。 Objective To study the expression of B7-H4 in primary epithelial ovarian cancer(EOC)and its clinicopathological significance.Methods The expression of B7-H4 mRNA and protein was detected in 86 cases of primary EOC(EOC group),40 cases of benign ovarian neoplasms(benign group)and 10cases of normal ovarian tissues(control group)by RT-PCR and immunohistochemistry PV-9000 2-step method,and the relationship between B7-H4expression and the clinicopathological parameters were analyzed.Results B7-H4 mRNA were detected in all groups,and the differences in optical density were statistically significant among the 3 groups(P〈0.05).The cytoplasmic and/or membranous staining of B7-H4 was detected in EOC with a positive rate of 77.9%(67/86),which significantly higher than that in benign group(5.0%,2/40) and control group(0%,0/10)(P〈0.05).There was no correlation between B7-H4protein expression and histopathological types,clinical stage,pathological grade,age,ascites cytology and lymph node metastasis(P〉0.05).Conclusion The high expression of B7-H4 in EOC indicates that B7-H4 plays an important role in cancer generation and should be further investigated as a potential diagnostic marker and therapeutic target for ovarian cancer.
