氧化苦参碱对内毒素作用下人牙周膜细胞增殖、细胞周期和超微结构的影响 被引量:5

摘要 目的研究氧化苦参碱(OMT)对内毒素(LPS)引起人牙周膜细胞(PDLC)增殖、细胞周期和超微结构的影响。方法体外分离及培养PDLC,实验组分别为LPS和不同浓度的OMT溶液的不同组合,对照组为仅含1%FBS的DMEM培养液。采用MTT法检测PDLC的增殖情况;流式细胞术测定PDLC的周期分布,透射电镜下观察细胞超微结构的改变。结果 LPS能够明显抑制PDLC的生长与增殖,流式细胞测定DNA合成前期细胞比例明显上升,而DNA合成期细胞比例下降,对PDLC超微结构有明显的损伤,OMT可改善此现象。结论 OMT可在一定程度上阻断LPS对PDLC的破坏,对PDLC有保护作用。 Purpose To observe the effects of oxymatrine on multiplication situation and cell cycle and ultrastructure of PDLC under existing LPS.Methods Firstly,detach PDLC externally and cultivate them,for oxymatrine administration group different combinations of LPS and oxymatrine in different concentrations are used,and for the other group DEME nutrient fluid of 1% FBS is used.Then,MTT method was used to examine PDLC′ multiplication situation.Flow cytometry was used to measurate the cycle distribution of PDLC.Transmission electron microscope(TEM)was used to observe the change of PDLC ultrastructure.Results LPS can significantly inhibit the proliferation of PDLC.The percentage of human periodontal ligament fibroblast in G1 phase during exponential growth was increased,while the percentage of human periodontal ligament fibroblast in S phase was decreased.It had significant damage to the ultrastructure of PDLC,but oxymatrine could ameliorate above phenomenas.Conclusion Oxymatrine can interrupt LPS′s damage to PDLC to some extent,and provide protection to PDLC.
