摘要
目的探讨夏枯草提取物对碳水化合物水解和葡萄糖吸收影响的作用机制。方法以Caco-2细胞为研究对象,分设对照组、阳性药阿卡波糖组(25μg/mL)、夏枯草水提物组(0.75μg/mL)、浸膏(0.25μg/mL)组、多糖(0.5μg/mL)组。采用RT-PCR法测定Caco-2细胞的α-葡萄糖苷酶、SGLT-1、GLUT-2、Na+-K+-ATP酶mRNA表达。结果夏枯草提取物对Caco-2细胞α-葡萄糖苷酶、SGLT-1、GLUT-2、Na+-K+-ATP酶mRNA表达,与对照组比较均有显著性差异,阿卡波糖也有类似的抑制作用。结论夏枯草提取物通过抑制Caco-2细胞α-葡萄糖苷酶、SGLT-1、GLUT-2、Na+-K+-ATP酶mRNA表达作用,来延缓对碳水化合物水解和影响葡萄糖吸收,可能与降低餐后血糖水平有关。
Purpose To examine the mechanism by which Prunella vulgaris L.extract affects the carbohydrate hydrolysis and absorption of glucose.Methods Caco-2 cells were treated with or without the decoction of water of Prunella vulgaris L.(0.75 μg/mL),the sodden ointment of(0.25 μg/mL),and the sodden ointment of(0.5 μg/mL),respectively.Acarbose(25 μg/mL) served as positive control.mRNA expression of α-glucosidase,SGLT-1,GLUT-2,Na^+-K^+-ATPase were examined by RT-PCR.Results The data showed that Prunella vulgaris L.extract significantly decreased the expression of α-glycosidase,SGLT-1,GLUT-2 and Na^+-K^+-AT enzyme,respectively,in Caco-2 cells.Acarbose has a similar inhibitory effect.Conclusion Prunella vulgaris L.extract inhibits the α-glucosidase,SGLT-1,GLUT-2,Na^+-K^+-ATP enzyme mRNA expression in Caco-2 cells.The data indicate that Prunella vulgaris L.extract could reduce the blood sugar levels after meal through affecting the glucose hydrolysis and the absorption of carbohydrates.
