摘要
目的探讨三拗芎葶合剂对哮喘大鼠肺组织蛋白激酶C(PKC)、基质金属蛋白-9(MMP-9)的影响。方法 60只SD大鼠随机分为正常组、模型组、地塞米松组、三拗芎葶剂高剂量组(简称高剂量组)、三拗芎葶剂中剂量组(简称中剂量组)、三拗芎葶剂低剂量组(简称低剂量组),每组10只。除正常组,其余各组复制哮喘大鼠模型。造模成功后,正常组大鼠腹腔注射致敏用等体积0.9%氯化钠注射液代替卵清白蛋白(OVA)氢氧化铝混合液,超声雾化吸入激发用等体积0.9%氯化钠注射液代替OVA。各组均于激发前30 m in予相应药物治疗,高剂量组、中剂量组、低剂量组均给予三拗芎葶合剂(分别按照20、10、5 g/kg给药)4 mL,每日1次灌胃;地塞米松组给予醋酸地塞米松注射液(按照1 mg/kg给药)2 mL,每日1次腹腔注射;正常组和模型组均给予4 mL0.9%氯化钠注射液灌胃。给药2周后处死大鼠,取右肺组织,观察各组大鼠肺组织病理形态学变化及支气管黏膜和黏膜下嗜酸粒细胞数(EOS)和淋巴细胞数;检测各组大鼠支气管、肺组织PKC及MMP-9含量。结果肺组织苏木精-伊红(HE)染色观察发现,模型组支气管黏膜下层和平滑肌层较对照组明显增厚,管腔变形、狭窄,肺泡膈明显增宽,可见血管增生和扩张,气道壁和肺组织中可见以嗜酸性粒细胞、淋巴细胞为主的大量炎症细胞浸润;各治疗组较模型组有所改善,高、中剂量组及地塞米松组优于低剂量组(P<0.05)。模型组EOS和淋巴细胞计数均明显高于正常组(P<0.05),地塞米松组、高剂量组、中剂量组及低剂量组EOS和淋巴细胞计数均低于模型组(P<0.05),低剂量组和地塞米松组EOS和淋巴细胞计数差异均有统计学意义(P<0.05),低剂量组均高于地塞米松组。模型组PKC和MMP-9阳性反应产物平均光密度值均明显高于正常组(P<0.05),地塞米松组、高剂量组、中剂量组及低剂量组PKC及MMP-9阳性反应产物平均光密度值均低于模型组(P<0.05),低剂量组PKC及MMP-9阳性反应产物平均光密度值较地塞米松组明显升高(P<0.05)。结论三拗芎葶合剂可明显降低大鼠气管黏膜和黏膜下EOS和淋巴细胞数,改善肺组织的病理变化,减弱肺组织PKC和MMP-9表达。
ObjectiveTo investigate the effect of Sanaoxiongting misture on the contents of PKC and MMP-9 in lung tissue of rat with asthma.Methods 60 SD rats were randomly divided into six groups,control group,model group,high-,middle-,low-dose Sanaoxiongting misture group and Dexamethasone group,10 rats in each group.Besides control group,rat model of asthma were established in other groups.Equivalent volume 0.9% NaCl instead of mixture of OVA and aluminium hydroxide for sensitization was applied in control group.Ultrasonic nebulization inhalation of equivalent volume 0.9% NaCl instead of OVA for suscitation was applied in control group.30 min utes before suscitation corresponding medication was carried on in every treatment groups.Sanaoxiongting misture 20 g/kg,10 g/kg and 5 g/kg 4 mL were lavaged in high-,middle-,low-dose Sanaoxiongting misture group.Dexamethasone 1 mg/kg was intraperitoneal injected in Dexamethasone group.After two weeks,changes of pathomorphology in lung tissue and EOS and lymphocyte in bronchial mucosa and submucous were observed.PKC and MMP-9 in bronchus and lung tissue were detected.Results Bronchial submucous and smooth muscle layer in model group was thickened in comparison with that in control group.The alveolar diaphragm was widening,inflammatory cell infiltration were observed in airway wall and lung tissue.Improvement of inflammatory cell infiltration was observed in each treatment groups.The improvement of inflammatory cell infiltration in high-,middle-dose group and Dexamethasone group was superior to that in low-dose group(P〈0.05).EOS and lymphocyte in model group was higher than those in control group(P〈0.05).EOS and lymphocyte in high-,middle-,low-dose group and Dexamethasone group was lower than those in model group(P〈0.05).And EOS and lymphocyte in low-dose group was higher than those in Dexamethasone group(P〈0.05).PKC and MMP-9 in model group was higher than those in control group(P〈0.05).PKC and MMP-9 in high-,middle-,low-dose group and Dexamethasone group was lower than those in model group(P〈0.05).And PKC and MMP-9 in low-dose group was higher than those in Dexamethasone group(P〈0.05).Conclusion Sanaoxiongting misture can decrease obviously EOS and lymphocyte in tracheal mucosa and submucous,improve the pathological changes in lung tissue and reduce the expression of PKC and MMP-9.
