摘要
目的探讨维拉帕米对裸鼠胶质瘤U251模型卡氮芥(BCNU)疗效的影响。方法制作裸鼠胶质瘤U251模型,分别给予BCNU和BCNU+维拉帕米进行治疗,测量肿瘤大小,用免疫荧光染色检测肿瘤中P-糖蛋白(P-gp),多药耐受蛋白1(MRP1)和肺耐药相关蛋白(LRP)的表达。结果 BCNU+维拉帕米组中,肿瘤大小明显比BCNU组减小,尤其是第一至四周更加明显(P<0.05)。肿瘤细胞中LRP,P-gp和MRP1均为阳性,主要在细胞膜表达,从形态学上两组差别并不明显。结论维拉帕米在体内有明显增加胶质瘤U251对BCNU的敏感性,逆转耐药的作用。
Objective To approach the effects of verapamil on carmustine(BCNU) chemotherapy to U251 glioma in mice. Methods The U251 glioma athymic mouse models were made,and then treated with BCNU and verapamil + BCNU respectively. Expression of multidrug resistance 1(MDR1,P-gp) ,multidrug resistance protein 1(MRP1) ,and lung resistance-related protein(LRP) were determined by immunofluorescence staining. Results Compared with BCNU group,the tumors reduced significantly in verapamil + BCNU group,especially in the first to forth weeks(P 0.05) . The expression of P-gp,MRP1,and LRP were positive in U251 glioma cells,and had no changes in morphology. Conclusion Verapamil can increase U251 glioma cells' chemosensitive to BCNU,and reverse multidrug resistance.
