溴隐亭对MetS-PCOS大鼠胰岛素抵抗和氧化应激作用研究 被引量:1

摘要 目的观察溴隐亭对代谢综合征-多囊卵巢综合征(MetS-PCOS)大鼠胰岛素抵抗和氧化应激作用的影响。方法采用十一酸睾酮加绒促性素(HCG)建立MetS-PCOS大鼠模型,连续口服给予溴隐亭、二甲双胍、罗格列酮6周,观察大鼠体重、空腹血糖(FBG)、口服葡萄糖后2小时血糖(OGTT-2hBG)、胰岛素、肿瘤坏死因子-α(TNF-α)、睾酮(T)、丙二醛(MDA)、一氧化氮(NO)等的含量,以及超氧化物歧化酶(SOD)和诱导型一氧化氮合酶(iNOS)、一氧化氮合酶(NOS)的活性,并计算胰岛素敏感指数(ISI)的变化。结果 MetS-PCOS大鼠体重,FBG,OGTT-2hBG,胰岛素水平,TNF-α,T,MDA,NO和iNOS均明显升高,而ISI和SOD显著降低,与空白对照组比较,差异有高度显著性(P<0.01);溴隐亭、二甲双胍、罗格列酮均能不同程度地改善MetS-PCOS大鼠的上述病理改变(P<0.01或0.05)。结论溴隐亭可以拮抗MetS-PCOS大鼠胰岛素抵抗(IR)、增强胰岛素敏感性、纠正高胰岛素血症、高雄激素血症以及改善氧化应激状态。 Objective To investigate the effects of bromocriptine on insulin resistance and oxidative stress in rats with polycystic ovary syndrome and metabolic syndrome(MetS-PCOS) . Methods The model of MetS-PCOS rats induced by testosterone undecanoate and human chorionic gonadotropin(HCG) ,afterwards bromocriptine,rosiglitazone and metformin were taken by intragastric administration for 6 weeks. At the end of the experiment,the bodies weight of rats were measured,and the contents or activities of fasting blood-glucose(FBG) ,2-hours blood glucose after oral glucose tolerance test(OGTT-2hBG) ,insulin,tumor necrosis factor(TNF-α) ,testosterone(T) ,resistin,malondialdehyde(MDA) ,nitric oxide(NO) ,inducible nitric oxide synthase(iNOS) and nitric oxide synthase(NOS) were determined,and the changes of insulin sensitivity index(ISI) were counted. Results Compared with the normal group,the bodies weight of rats and concentrations or activities of FBG,OGTT-2hBG,insulin,TNF-α,T,resistin,MDA,NO and iNOS increased obviously(P 0.01) ,but ISI and SOD decreased significantly(P 0.01) . All of drugs could improve the pathologic changes in MetS-PCOS rats(P 0.01 or P 0.05) . Conclusion Bromocriptine is able to antagonist insulin resistance,enhance the insulin sensitivity,modify hyperinsulinemia,hyperandrogenemia and oxidative stress state in MetS-PCOS rats.
