摘要
采用单因素和正交试验设计,通过摇瓶培养对链霉菌菌株F-15产生抑菌活性物质的发酵条件进行优化。单因素试验结果表明,其最佳发酵条件为:玉米粉作碳源,KNO3作氮源,初始pH为7,培养时间为5 d,250 mL的摇瓶装液量为60 mL,接种量为150μL。优化后的培养基配比为:玉米粉3.0%、KNO33.0%、MgSO40.2%、NaCl 0.2%。
The fermentation conditions of the antagonistic substances of the Streptomyces strain F-15 produced in shake flasks were optimized by using the method of single factor and orthogonal experimental design. The results showed that the optimal carbon and nitrogen sources were corn flour and KNO3. Initial pH 7 of medium, culture for 5 days, 60 mL liquid volume in 250 mL shake flask, and an inoculum size of 150μL were optimal fermentation conditions. Based on orthogonal experiments, the best culture medium composition was 3.0% corn flour, 3.0% KNO3, 0.2% MgSO4 and 0. 2%NaCl.
