摘要
目的对进展期与好转期结核性脑膜炎患者的脑脊液进行差异表达的蛋白分析,鉴定与结核性脑膜炎相关的蛋白质。方法取进展期与好转期结核性脑膜炎患者的脑脊液,利用双向电泳技术分离脑脊液全蛋白,软件分析找出差异表达蛋白,再利用基质辅助激光解吸/电离串联飞行时间质谱技术对差异表达的蛋白质进行分析。结果进展期与好转期结核性脑膜炎患者脑脊液总蛋白的双向电泳图谱产生差异的点有68个,蛋白质相对分子质量(10~100)×103,等电点值为pl3~pl10;鉴定出3个与结核性脑膜炎相关的蛋白质,分别为超氧化物歧化酶、视黄醇结合蛋白及38×103糖脂蛋白。结论 3个差异表达的蛋白质有可能作为结核性脑膜炎诊断的候选标志物、治疗或疫苗候选物,进行深入研究。
OBJECTIVE To identify the different expression of proteins in the cerebrospinal fluid collected from progressive and recovery stages of tuberculous meningitis patients.METHODS Two-dimensional electrophoresis was used to compare the global protein patterns in the cerebrospinal fluid between the two stages,MALDI-MS/MS was used to identify the different expression of proteins.RESULTS From two dimensional electrophoretic maps of whole proteins of the cerebrospinal fluid between the two stages,68 different expressed protein spots were found,and their relative molecular mass and isoelectric point ranged from 10×103 to 100×103 and from pI 3 to pI 10,respectively.Three protein spots in gel were identified as superoxide dismutase(SOD),retinol-binding protein and 38×103 glycolipoprotein.CONCLUSION These 3 proteins may be regarded as signs of tuberculous meningitis diagnosis and the candidates for treatment or vaccine,which are also very important for the further study.
