摘要
研究大孔树脂分离纯化苹果类黄酮工艺。采用AB-8,DM-130,NAK-9三种大孔树脂对苹果类黄酮进行吸附解析,以类黄酮吸附和解析率为指标综合评价,优选出最佳树脂;对优选出的树脂进行吸附和解析条件探讨。结果表明:3种大孔树脂吸附率分别为70.55%,62.45%,74.13%,解析率分别为92.66%,86.84%,87.22%。AB-8树脂综合性能较好。优化AB-8树脂吸附工艺参数为:吸附液浓度0.64 mg/mL吸附液pH值6.0,吸附流速1 mL/min。解析工艺参数为:解析乙醇体积分数70%,解析流1.6 mL/min。苹果类黄酮回收率及纯度分别可达89.46%,为92.50%。
To establish a technical procedure for the purification of flavonoids from apple. Three types of macro porous resin (AB-8, DM-130, and NAK-9) were used to absorb and desorb apple flavonoid. The rate of absorption and desorption were compared among the resins. The conditions of absorbability and desorption were further investigated. The absorbability rate of three types of macro porous resin were 70.55% ,62.45% ,74. 13% respectively, The desorption rate were 92.66% ,86.84%. 87.22% respectively. The results indicated that AB-8 macroporous resin was the best. The optimum adsorption conditions were as follows: sample concentration 64 mg/mL, pH6.0 and the feeding rate 1 mL/ min. The optimum desorption parameters were as follows: ethanol concentration 70% ,the flow rate. 1.6 mL/min. The reclaimed ratio was 89.46% and the purity of the flavonoids extracted from apple fruit was 92.5%.
