自制复方黄芪汤治疗慢传输型便秘与C—kit结肠组织表达的研究 被引量:2

Self-designed Compound Astragalus Decoction for Treating Slow Transit Constipation and C-kit Expression in Colon Tissue: An Experimental Study

摘要 为研究结肠慢传输型便秘(STC)大鼠结肠组织Cajal间质细胞(ICC)的分布、形态和数量变化,以及与大鼠结肠组织中CkitmRNA的表达的关系,观察自制汤剂复方黄芪汤治疗STC模型大鼠的疗效,将大鼠32只随机分为两组:模型组(24只)和生理盐水对照组(8只),模型组饲料中添加大黄粉制作便秘模型,生理盐水对照组饲以普通软饲料。停药一周后从模型组中随机选取8只,与生理盐水对照组同时禁食不禁水24h后,测活性炭推进率,处死,取出标本。测模型组结肠组织中ICC的数量和形态改变,以及组织中C—kitmRNA的表达改变,并与生理盐水对照组进行比较。另外16只随机分为生理盐水空白组(8只)和中药干预组(8只),中药干预组灌服自制汤剂复方黄芪汤10ml/(kg·d),约含生药2g/ml。灌药30d。生理盐水空白组灌服同体积的生理盐水30d,测活性炭推进率,处死,取出标本。测中药干预组结肠组织中ICC的数量和形态改变以及组织中C—kitmRNA的表达改变,并与生理盐水空白组进行比较。结果显示,与生理盐水对照组比较。模型组炭末推进长度小于生理盐水对照组(P〈O.05),炭末推进率明显低于生理盐水对照组(P〈O.01)。Cajal间质细胞的形态变短、变钝,细胞数量变少(P〈O.05)。Ckit mRNA的表达量变少。与生理盐水空白组比较,中药干预组炭末推进长度大于生理盐水空白组(P〈0.05),炭末推进率明显高于生理盐水空白组(P〈0.01)。ICC形态恢复正常,细胞数量增多(P〈0.05)。C—kitmRNA的表达量变多(P〈0.05)。结果表明,大黄饲喂法造模是成功的,STC大鼠结肠组织中存在ICC的形态变短、变钝,细胞数量变少,CkitmRNA的表达量变小。自制汤剂复方黄芪汤能使STC大鼠结肠组织中ICC形态和数量向正常转化,使C—kitmR—NA的表达量增多。 In order to investigate the distribution,morphology and quantity changes of interstitial cells of Cajal(ICC) in colon tissue of rat with slow transit constipation (STC) ,and their relation with C-kit mRNA expression,authors observed the therapeutic effects of self-designed compound Astragalus decoction for treating STC of rat-model. They divided 32 rats randomly into two groups:model group ( n =24) and nor- mal saline solution controlled group( n =8,also named as controlled group). The animals of model groups eat the feed supplied with the powder of Rhubarb and controlled group supplied common feed respectively. Later one week after stopping use of above drug 8 rats were selected from model group randomly, along with rats of controlled group were ordered alone not to take food but may drink water for 24-hour,then the movement ratio of activated charcoal in rats colon tissue were detected,the rats were sacrificed for sampling their specimen. The changes of quantity, morphology of ICC, and C-kit mRNA expression were detected, then compared with that of controlled group; moreover 16 rats were divided randomly into two groups: blank controlled group(8 rats,only with normal saline solution) and intervention group (8 rats,with tradi- tional Chinese drugs). The rats of intervention group were irrigated with above self-designed decoction, 10ml/kg(about containing crude drugs 2g/ml),for 30 days; while the rats of blank controlled group, with same volume of normal saline solution,for 30 days;the specimen of both groups were taken out following detecting movement ratio of activated charcoal and sacrificeding animals. The changes of quantity,morphol- ogy of ICC of intervention group,and their C-kit mRNA expression were detected,and compared with that of blank controlled group. As results, in model group the movement length of activated charcoal particles was less than that in controlled group( P d0.05), and the movement ratio was significantly lower than that of controlled group( P d0.01) ,on ICC,their morphology became shorter and duller,and quantity Was less ( P (0.05),the expression of C kit mRNA also less^in intervention group the movement length was larger than that in blank controlled group( P %0.05), the movement ratio was significantly higher than that in blank controlled group( P (0.01), on ICC, their morphology recovered normal,and quantily also increased( P %0.05) ,expression of C kit mRNA increased ( P %0.05). Above results show that establis- hing model with the powder of Rhubarb is successful^the ICC in colon tissue of STC rat become shorter and duller morphologically, and its quantity decreases ~ the self-designed decoction can make the morphology and quantity of ICC convesing normal, the expression level of C-kit mRNA increasing.
