基于Flash GC E-Nose的鳀鱼酶解液吸附脱腥后树脂洗脱条件优化的初步研究

摘要 通过非极性大孔吸附树脂DA201-C处理鳀鱼酶解液,采用不同的洗脱条件对树脂柱进行洗脱,应用FlashGCE-Nose测定了标准液和不同洗脱液的腥味值,绘制出线性拟合曲线,比较了五种洗脱条件对鳀鱼酶解液树脂吸附样品的洗脱效果,并确定了最优洗脱条件。结果表明:不同组成和浓度的洗脱液对总氮和腥味物质的洗脱效果有所差别,乙醇洗脱液的洗脱效果要明显好于稀酸碱洗脱液。梯度乙醇洗脱液的总氮洗脱率最高,为96·80%;50%乙醇洗脱液的腥味值最高,达到4·65,腥味值和总氮洗脱率无明显的正相关关系。综合考虑这两个指标,选择梯度乙醇洗脱液进行树脂洗脱操作。 The enzymatic hydrolysate of anchovy was treated by non-polar macroporous adsorption resin DA201-C,the column of resin was desorbed by different eluent.Fishy odor of standard and eluent sample was detected by the improved method of Flash GC E-Nose,the curve of standard linear fit was drawn and five elution conditions were compared.Results showed that:eluting effects of total nitrogen and fishy odor substance were different under different conditions,ethanol-containing eluent had more obvious effect than the diluted-acid-and-alkali eluent.The total nitrogen desorption rate was the highest in gradient ethanol eluent,which was 96.80%,whereas the 50% ethanol eluent had the highest value of fishy odor,which was 4.65.Considering these two indexes,gradient ethanol eluent was chose as the best eluent.
