缬沙坦联合川芎嗪对血管性痴呆大鼠血浆6-keto-PGF_(1α)和TXB_2含量的影响 被引量:2

摘要 目的:检测血管性痴呆(VD)大鼠血浆6-keto-PGF1α和TXB2含量,比较缬沙坦、川芎嗪单独及联合对血管性痴呆(VD)大鼠学习记忆的改善作用,并分析原因。方法:反复夹闭双侧颈总动脉加硝普钠腹腔注射复制血管性痴呆大鼠模型;Morris水迷宫检测VD大鼠学习记忆力;放射免疫法检测大鼠血浆6-keto-PGF1α和TXB2含量。结果:与假手术组比较,模型组大鼠逃避潜伏期明显延长(P<0.01),跨越虚拟平台的次数、在平台象限停留的时间及平台象限游程百分比明显减少(P<0.01);血浆中6-keto-PGF1α含量降低,TXB2含量明显增加(P<0.01)。与模型组比较,缬沙坦、川芎嗪单独及联合均可缩短大鼠逃避潜伏期(P<0.05,0.01),增加大鼠跨越虚拟平台的次数、在平台象限停留的时间及平台象限游程百分比(P<0.05),三用药组间相互比较,以联合用药组效果更为明显(P<0.05);缬沙坦、川芎嗪单独及联合均可提高VD大鼠血浆6-keto-PGF1α含量,降低TXB2含量(P<0.05,0.01),三用药组间比较,联合用药对TXB2和6-keto-PGF1α的影响较缬沙坦明显,对6-keto-PGF1α的影响较川芎嗪明显(P<0.05)。结论:缬沙坦、川芎嗪单独及联合均可改善血管性痴呆大鼠的学习记忆功能,以联合用药效果更明显。其原因与其单独或联合用药均可提高VD大鼠血浆6-keto-PGF1α含量,降低TXB2含量,纠正VD大鼠循环血中TXA2/PGI2的平衡失调有关。 Objectives:To compare with effect of valsartan and ligustrazine given separately or in association on ability of learning and memory in rat with vascular dementia by detecting plasma 6-keto-PGF1α and TXB2 of VD rats,and to analyze the reasons.Methods:The vascular dementia models were duplicated by blocking bilateral common carotid artery of rat time and again and peritoneal injection sodium nitroprusside.The ability of learning and memory in rats were measured with morris water maze,and the plasma 6-keto-PGF1α,TXB2 was measured by radioimmunoassay.Results:Compared with the sham operation group,the rats in model group show longer escape latency(P0.01),and numbers of rats crossing platform,the time spent in target quadrant and the percentage of the distance spent in target quadrant obivously decreased(P0.01).The plasma 6-keto-PGF1α decreased,TXB2 were increased significantly(P0.01).Compared with model group,valsartan and ligustrazine given separately or in association can shorten escape latency(P0.05,0.01),numbers of rats crossing platform increased and the time spent in target quadrant and the percentage of the distance spent in target quadrant(P0.05),compared amongthe three treatment group,it was more obvious in combination treatment group(P0.05).Valsartan and ligustrazine were given separately or in association could decrease the plasma 6-keto-PGF1α and TXB2 levels(P0.05,0.01).Compared among the three treatment group,effect of valsartan united ligustrazine on the 6-keto-PGF1α is more obvious than valsartan or ligustrazine given separately,and on TXB2 is more obvious than ligustrazine given separately(P0.05).Conclusions:Valsartan and ligustrazine given separately or in association can both improve of ability of learning and memory in rats with VD,and the effect is more effective in the combination therapy group.The reason has to do with rectification dysequilibrium of TXA2/PGI2 in circulation of blood by enhancing plasma 6-keto-PGF1α and reducing TXB2 in rats.
