摘要
用本实验室筛选得到的一株黑曲霉以芦丁为底物进行液态发酵,以达到将芦丁转化为槲皮素的目的.采用正交实验法对液态发酵培养基及发酵条件进行优化,得到最佳发酵条件为:芦丁3%,NaNO_3 0.3%,K_2HPO_40.1%,MgSO_40.05%,KCl0.05%,FeSO_40.01%;pH=6;装液量20%,接种量20%,培养条件为35℃,200 r/min.在此条件下,芦丁的转化率为97.78%,槲皮素的得率为90.39%.采用正交实验法优化槲皮素的提取条件,得到最佳提取条件为酒精浓度60%,料水比1:6,温度60℃,提取4次,每次60min.在此条件下,槲皮素的提取率为95.37%.对提取得到槲皮素粗品进行热水沉淀,最终得到纯度为97.12%的槲皮素.
An Aspergillus niger had been screened to transformate rutin to quercetin in liquid fermentation. Optimizating the liquid fermentation medium and conditions by orthogonal test, the best conditions are as follows: rutin3%, NaNO3 0.3%, K2HPO4 0.1%, MgSO4 0.05%, KCl 0.05%, FeSO4 0.01%, pH=6; liquid volume 20%, inoculum 20%, temperature 35℃, shaker speed 200 r/min. In these conditions, the rate of tranfomation was 97.78 %, and the yield of quercetin was 90.39 %. Optimizating the extraction conditions of quercetin by orthogonal layout, the best extraction conditions are as follow: alcohol concentration 60%, the ratio of water and leavening 1 : 6, temperature 60℃, 4 times, and each for 60main. In these conditions, the extraction rate of quercetin was 95.37 %. Refining the quercetin with hot water, the final purity of purifier quercetin was 97.12%.
