温度、pH及碳氮源对产色素青霉菌株(Penicillium sp.)生长的影响 被引量:2

Effects of pH,Carbon Sources and Nitrogen Sources of Mediums and Incubation Temperature on the Growth of a Penicillium Strain

摘要 PSO312是从土壤分离的1个青霉菌株,生长中能产生不同酸碱性条件下显黄色与紫红色等天然色素物质.通过菌株固体平板上恒温培养,168 h测定菌落直径的方法研究了温度、pH及碳、氮源对菌株(Penicillium sp.)生长的影响.研究表明31℃为菌株生长的最适温度.31℃培养以pH 2~3的最快,pH 8~9生长次之,24℃培养以pH 8~9生长最快,pH3~4其次.菌株以蛋白胨4 g/L、牛肉膏4 g/L、酵母膏2g/L及尿素2 g/L为氮源培养的菌落直径分别为55.75 mm、51.5 mm、55.5 mm及25.3 mm.在(NH4)_2SO_4 2g/L、NH_4NO_33g/L、NaNO_3 4g/L、KNO_3 4g/L用量之上培养生长最快,菌落直径分别为35 mm、34mm、35 mm、及32 mm.葡萄糖、蔗糖用量30 g/L、果糖20 g/L及乳糖25 g/L的生长速率最快,菌落直径分别为45 mm、38 mm、38 mm及39 mm.丙三醇与淀粉25 g/L培养的菌落直径均为39 mm. PSO3 12 (Penicillium sp.) strain isolated from a soil, can produce yellow and purple natural pigments cultured with different pH media. The effects of temperature, pH value, Carbon sources and nitrogen sources on the strain growth was investigated according to the colony diameter after which was cultured on the agar plate media for 168h. The results showed that the strain growth optimal temperature was 31 ℃. The strain cultured at 31 ℃ grew the fastest on pH 2-3 media, the second on pH 8~9 media. And then the strain cultured at 24 ℃ grew the fastest on pH 8~9 media, the second on pH 3~4 media. The colony diameters cultured on the media contained peptone 4 g/L, beef extract 4 g/L, yeast extract 2 g/L and urea, 2 g/L as organic nitrogen sources were 55.75 mm, 51.5 mm, 55.5 mm and 25.3 mm respectively. The colony diameters cultured on the media contained (NH4)2SO4 2 g/L, NH4NO3 3 g/L, NaNO3 4 g/L and KNO3 4 g/L as inorganic nitrogen sources were 35 ram, 34 mm, 35 mm and 32 mm respectively. The colony diameters cultured on the media contained glucose and sucrose 30g/L, fructose 20 g/L and lactose 25 g/L of the fastest growth rate, colony diameters of 45 ram, 38 mm, 38 mm and 39 mm. The strain on the dosages of glycerol and starch 25 g/L grew the fastest respectively.
