冬虫夏草对糖尿病大鼠肾组织骨桥蛋白表达的影响 被引量:6

摘要 目的:探讨冬虫夏草对糖尿病大鼠肾组织中骨桥蛋白(OPN)表达的影响。方法:将Wistar大鼠建成链脲佐菌素诱导的糖尿病模型。设对照组(VH组);糖尿病肾病组(DN组);冬虫夏草治疗组(CS组)。16周后观察各组大鼠血糖、血肌酐、24h尿蛋白排泄量、肾组织血管紧张素(Ang)Ⅱ含量等指标及肾间质纤维化评分,并用免疫组织化学方法和Northern blotting法分别观察肾组织OPN、单核巨噬细胞表面特异性标志抗原(ED-1)的表达及OPNmRNA的活性。结果:冬虫夏草能降低血糖、血肌酐、24h尿蛋白排泄量、肾组织AngⅡ含量,减轻肾间质纤维化,减少肾组织OPN的表达、OPNmRNA的活性;同时间质单核巨噬细胞浸润显著减少,且与肾小管骨桥蛋白的表达呈正相关。结论:冬虫夏草的肾脏保护作用可能与下调肾组织OPNmRNA活性介导的间质巨噬细胞的浸润有关。 Objective:To observe the effect of Cordyceps(CS) on osteopontin(OPN) expression in rats with diabetic nephropathv.Methods:Wistar rats were randomized into three groups: VH group(sham group);DN group(untreated diabetic group);CS group(CS treated diabetic group).Diabetic rats were induced by intraperitoneal streptozotoeia iniection(50mg/kg).The changes of blood glucose(BG),serum creatinine(Scr),urinary protein excretion(ALB-n),tubulointerstitial fibrosis score(TFS),renal angiotensin(Ang) Ⅱ were evaluated after 16 weeks.In addition,immunohistochemical method and Northern blotting analysis were used to detect OPN immunoreactivity,ED1-positive monocyte/macrophage and OPN mRNA expression.Results:CS significantly decreased BG,Scr,AIB-n,TFS,levels of Ang Ⅱ.CS could inhibit ED1-positive cell infiltration,accompanied with down-regulation of OPN immunoreactivity and OPNmRNA expression.Conclusion:CS could anenuate diabetic nephropathy,and this may be associated with reducing monocyte/macrophage infiltration induced by OPN in renal.
