一种胚胎体外培养的新方法:滴中孔培养法 被引量:1

A New Method for Embryo Culture in vitro: the Well in the Drop (WID) System

摘要 目的:建立并优化一种胚胎体外培养的新方法——滴中孔培养法。方法:用圆头玻璃管在培养皿上烫出多个圆孔,作为滴中孔培养的专用平皿;收集牛卵丘-卵母细胞复合体(COCs)并在滴中孔培养专用平皿中进行体外成熟(IVM)、体外受精(IVF)和体外培养(IVC)。结果:①滴中孔培养法与群卵共培养的效果相似,而单卵培养明显降低了卵母细胞的发育能力(P<0.05)。②培养滴体积和培养密度对卵母细胞核成熟没有明显的影响,但显著影响胚胎的发育能力。在培养密度不变的情况下,培养滴体积太小显著削弱了胚胎的发育能力,而培养密度过小或者过大也削弱胚胎的发育能力(P<0.05)。结论:滴中孔培养法可以达到群卵共培养的效果。 Objective: To establish and optimize a new method of embryo culture in vitro: the well in the drop (WID) system. Methods: The glass tube was used to make some round hole on the dish, to make a special dish for WID system; Bovine COCs and cultured in WID system in vitro maturation (IVM), in vitro fertilization (IVF) and in vitro culture (IVC). Results: 1) The WID culture system works as efficient as group culture, whereas the individual culture impaired the oocyte developmental competence significantly (P〈0.05). 2) The medium volume and density had no effect on oocyte nuclear maturation but affect embryonic development significantly. As to development in vitro, it was proved that too small medium volume was detrimental to embryo development with the same culture density. Besides, too low or too high culture density was proved detrimental to embryo development (P〈0.05). Conclusion: The WID culture system can achieve the effect of group culture.
