摘要 目的:观察壳聚糖温敏凝胶的微观形态、释放釉基质蛋白(EMPs)速率和大鼠体内组织相容性,以作为牙周组织再生支架材料的可行性进行初步研究。方法:制备壳聚糖温敏凝胶并通过扫描电镜观察其微观结构,用考马斯亮蓝试剂盒检测含EMPs浓度分别为150、100、50、25μg/mL的壳聚糖温敏凝胶释放速率;取6只SD大鼠随机分为3组,每组2只,将壳聚糖温敏凝胶注射入大鼠皮下,注射后第10、20、30天分别处死一组大鼠,HE染色法观察组织相容性。结果:壳聚糖温敏凝胶具有规则的多孔结构,含EMPs浓度150μg/mL组释放速率最快,25μg/mL组释放速率最慢,4组不同浓度的EMPs均可以在壳聚糖温敏凝胶中持续释放4周以上。HE染色观察显示壳聚糖温敏凝胶在大鼠体内具有较好的组织相容性和可吸收性。结论:壳聚糖温敏凝胶可以缓释EMPs并且在大鼠体内具有较好的组织相容性,作为支架材料应用于牙周组织再生具有广阔前景。 AIM:To investigate the micro-morphology,release rate of enamel matrix proteins ( EMPs) and histocompatibility of chitosan thermosensitive hydrogel,and to preliminarily evaluated the feasibility of chitosan thermosensitive hydrogel as a scaffold material in periodontal tissue regeneration. METHODS:Chitosan thermosensitive hydrogel was prepared and the micro-morphology was observed by scanning electron microscope. Chitosan thermosensitive hydrogels were loaded with 150 μg/mL,100 μg/mL,50 μg/mL and 25 μg/mL EMPs respectively and the releasing rate of EMPs was monitered by coomassieblue staining kit in vitro. Six rats were randomly divided into 3 groups with 2 rats in each group. Chitosan thermosensitive hydrogel was subcutaneously injected into the rat. At 10 d,20 d and 30 d after injection,one group was sacrificed. The histocompatibility was investigated by HE staining. RESULTS:Chitosan thermosensitive hydrogel showed a porous structure. The release rate of 150 μg/mL group was the fastest and the 25 μg/mL group was the lowest. The release of EMPs in each group lasted for more than 4 weeks. Chitosan thermosensitive hydrogel demonstrated histocompatibility and absorbability in rats. CONCLUSION:Chitosan thermosensitive hydrogel can release EMPs slowly and has a good histocompatibility in vivo and is likely to be used as a new scaffold material in the regeneration of periodontal tissue.
