微小RNA在肾脏及其相关疾病中的调控作用

摘要 微小RNA(miRNA)是细胞内源性的一种非编码的RNA,可表达出原始miRNA转录物(pri-miRNA),较大的primiRNA经一种RNA内切酶Ⅲ(Drosha)和辅助因子Pasha(含有双链RNA结合区域)加工成单链miRNA前体,随后具有发卡结构的单链miRNA前体在细胞质中被双链RNA专一性RNA内切酶(Dicer)切割成约22 nt的miRNA.成熟的miRNA中的一条单链与RNA诱导的沉默复合体(RNAinduced silencingcomplex,RISC)结合,形成RISC复合体.通过与靶基因mRNA的3'非翻译区(3'UTR区域)的碱基完全或不完全配对,导致mRNA降解或mRNA翻译抑制,即转录后基因沉默[1].
