摘要
从淀粉厂附近的酸性土壤、酒醅、醋渣等样品中筛选到了7株产酸性葡聚糖水解酶的微生物菌株,通过复筛发现醋渣来源的B-1-1菌株产生的酸性葡聚糖水解酶活性较强.初步鉴定B-1-1为嗜热脂肪芽孢杆菌.对该菌株的产酶条件及酶学特性进行了研究,结果为:在优化后的培养基上,37℃,220r/min摇床培养48h,产酶pH=5.0,温度为70℃时B-1-1产酶活性最高,可达185.3U/mL.
Seven microbial strains of producing acid glucan hydrolase were screened from acid soil,fermented grains and vinegar sauce.After re-screeing,the B-1-1 strain that came from vinegar sauce produced strong enzyme activity.The strain was indentified as Bacillus stearothermophilus.Fermentation conditions and enzyme characteristics showed that the optimal conditions were 37 ℃,220 r/min shaking culture 48 h,pH=5.0,temperature 70 ℃,enzyme activity of B-1-1 up to 185.3 U/mL.
