摘要
目的:通过观察"百会"透"曲鬓"头针疗法对急性脑出血大鼠脑源性神经营养因子(BDNF)、神经生长因子(NGF)表达的影响,揭示脑出血后神经重塑的可能机理。方法:通过自体血注入法复制急性脑出血模型,术后随机分为模型组、针刺组和西药组。运用原位杂交法检测各组大鼠脑出血后6h、1d、2d、3d、7d五个时相点BDNFmRNA和NGFmRNA的表达变化。结果:①各造模组大鼠血肿周围脑组织BDNFmRNA的表达随不同时相点的变化而逐渐增加,至2d时达高峰,3至7d时逐渐下降。在相同时相点,针刺组BDNF的表达均明显高于模型组和西药组,且差异有统计学意义(P<0.01)。②各造模组大鼠血肿周围脑组织NGFmRNA的表达随不同时相点的变化而逐渐增加,至7d时达高峰。在相同时相点,针刺组NGF的表达均明显高于模型组和西药组,且差异有统计学意义(P<0.01)。结论:"百会"透"曲鬓"头针疗法可能是通过增加内源性BDNFmRNA和NGFmRNA的合成与分泌,以促进脑出血损伤后神经的修复与重塑,从而实现对出血后继发性脑损伤的神经保护作用。
Objective:To observe BaiHui-QuBin scalp-acupuncture on BDNF and NGF expression in intracerebral hemorrhage(ICH) rats and to explore the possible mechanisms of nerves remodeling.Methods:The ICH rat models were established by.The rats were assigned into 3 sets randomly:the model set,the pinprick set and the western medicine set.We detected the positive expression of BDNFmRNA and NGFmRNA at 6h,1d,2d,3d,7d after ICH in situ hybridization technique.Results:①All of the rats after being made model,the expression of BDNFmRNA of rat's brain tissue surrounding the hematoma was increased gradually and to the peak in 2days with the increasing of different time phase point,but decreased gradually in 3days and 7days.In the same time phase point,compare with the model set and the western medicine,the pinprick set's expression of BDNFmRNA was higher obviously and had the statistical significant(P0.01).②All of the rats after being made model,the expression of NGFmRNA of rat's brain tissue surrounding the hematoma was increased gradually and to the peak in 7days with the increasing of different time phase point.In the same time phase point,compare with the model set and the western medicine,the pinprick set's expression of NGFmRNA was higher obviously and had the statistical significant(P0.01).Conclusions:The therapy of BaiHui-QuBin scalp-acupunctur possibly through the way of promoting the expressing of endogenous BDNFmRNA and NGFmRNA and bring into full play the role of nerves remodeling in acute intracerebral hemorrhage.
